Ampelomyces quisqualis

Isolation and culturing methods:

Isolated from soil located in the Bucknell University Greenhouse, on the 4th floor of the Biology building. Sample was placed on malt extract agar + chloramphenicol and allowed to grow for a week. Pure sample was obtained by sectioning a small sample of the growth onto MEA plate.

 

Culture appearance and growth:

Surface texture appears white and fuzzy 1wk into growth. Small black pores (>1mm in diameter) appear after 3wks of growth. Hyphae measure 4-5um wide, with septa present, but lacking clamp connections.

 

Spore production:

Cleistothecia are dark brown, measuring 62.5-67.5 um x 100-105um and develop on agar surface, not embedded. Spores are dikaryotic and oval shaped, measuring 2-3um.x 4-5um.

ETA: PCR sequencing and BLAST analysis suggest this speceies is Didymella americana, in the class Dothideomycetes.
