今日、 日本やドイツをはじめ世界各国でラガータイプのビールが醸造されてレヽ る。 このタイプのビール醸造に使用する酵母の多くは発酵後期に凝集し、 発酵夕 ンクの底部に沈降する性質を持つことから、 下面ビール酵母と呼ばれている。 こ の酵母の凝集性は、 ビール醸造工程において酵母回収やその後の濾過並びにビー ルの香味や品質に影響を及ぼす重要な性質である。 また、 下面ビール酵母は性質 が安定している場合、生産における優位性から回収酵母を繰り返し ί 用している。 そのため、 下面ビール酵母の凝集性を判定することは、 新たな酵母のスクリー二 ング ·選択などにおいて重要である。
酵母の一般的な凝集性測定法としては Burns法 (J.Inst Brew., 43, 31, 1937)、 Helm法 (Wallerstein Laboratory Communications 1£, 315, 1953)および数多く の改良法がある。 これらの方法はいずれも培養 ·発酵後の酵母を集菌し、 酵母を 凝集 ·沈降しやすい環境下 (すなわち pH4.5 前後、 低温) で、 凝集促進物質で ある Ca イオンと共存させることにより、 人工的に凝集性を発現させてその凝 集 ·沈降状況を測定する方法である。

発明の開示
本発明では発酵を行わず短期間で、 容易に、 再現性良く下面ビール酵母の凝集 性を評価する方法を確立することを目的とした。
実験室酵母サッカロマイセス ·セレピシェの第 νπι 染色体上に存在する凝集 性遺伝子 ^£Li2^の ORF配列 (Science, 2£5, 2077 (1994))を基に設計したプライ マ一を用いて、 PCR を実施し、 目的断片の増幅の有無およびそのサイズを調査 する,レにより、 さらに、 凝集性下面ビール酵母のゲノムを錶型とし、 上記記載 のプライマ一を用いて PCR を実施し、 増幅した産物をプローブとして、 サザン ハイブリダィゼ一シヨンを行うことによって、 下面ビール酵母の;疑集 ^fe有無の判 定および非凝集性化変異の検出が可能になることを見い出し、本発明を完成した。 すなわち、 本発明の第 1は実験室酵母サッカロマイセス ' セレピシェ ( accharomyces cerevisiae) の凝集性遺伝子 FLQ5の ORF配列を利用するこ とにより下面ビール酵母の凝集性有無を判定する方法である。
本発明の第 2は同配列を利用することにより本来凝集性を持つ下面ビール酵母 の変異による非凝集性化および凝集性低下を判定する方法である。
本発明の第 3は同配列を利用することにより本来凝集性を持つ下面ビール酵母 の変異による非凝集性化、 凝集性低下を予知する方法である。
本発明の第 4は同配列から設計したプライマーを用いて、 P C R反応を行い、 増幅産物の有無を検出することにより、 下面ビール酵母の凝集性有無を判定する 方法である。
本発明の第 5は同配列から設計したプライマーを用いて、 P C R反応を行い、 増幅産物の有無を検出することにより、 下面ビール酵母の変異による非凝集性化 および凝集性低下を判定する方法である。

以下に本発明を詳しく説明する。 判定対象となる酵母のゲノムの調製は公知の いかなる方法も使用可能である(例えば Methods in Yeast Genetics, Cold Spring Harbor Laboratory Press, p l30(1990))。 さらに、 多数の試料から調製する場合 は、 近年著しい進歩を遂げている自動 DNA抽出装置が容易で効率がよく有効で ある。
遺伝子の存在状況の確認は従来から知られているいずれの方法も取り得る。 以 下に主たる方法を示す。 ①判定対象となる酵母から抽出したゲノムを錡型として連続した 10bp以上の 塩基からなるプライマ一対を用いて PCR法によって核酸の増幅を行う。 安定し た高い検出感度を得るためにはプライマー長は約 20bp前後が好ましい。 また、 プライマーの組み合わせにあたり C末端側は 0〜約 2000bpの間の DNA分子あ るいはその塩基配列に対して相補的な塩基配列を含む DNA分子からなるプライ マーと、 N末端側は約 2400〜3228bpの間の DNA分子あるいはその塩基配列に 対して相補的な塩基配列を含む DNA分子からなるブラィマーと設計することが ポイントとなる。 プライマーの例を表 1 に示すが、 センス配列として C末端側 の配列を選択した場合、 アンチセンス側として任意の N末端側の配列を選択す ればよい。 すなわち、 プライマー例に示す、 a配列に対して、 c配列あるいは d 配列のいずれを選択しても判定は可能であり、 同様に b配列に対して、 c、 dい ずれの配列を選択して良い。 配列 aは配列表の配列番号 1に、 配列 bは配列番号 2に、 配列 cは配列番号 3に、 配列 dは配列番号 4に対応する。 上記の DNA分 子は公知の方法に従い、 化学合成することが出来る。 また、 専門業者に委託する ことも可能である。
PCR反応に用いるポリメラ一ゼ等の起源は問わないが、 耐熱性に優れ、 安定 して長鎖の増幅をおこなうことができるものが好ましい。 また、 プライマー対と 判定対象ゲノム以外があらかじめ混合されているキットの利用、例えば、 TaKaRa Perfect Shot (宝酒造製) などを利用することが容易に安定した結果を得るため に有効である。 反応条件は一般的な PCR条件でよく、 熱変性温度 90〜95°C、 ァニーリング温度 40〜65°C、 伸長温度 70〜75°C、 サイクル回数 20回以上が好 ましい。
得られた増幅産物は常法に従ってァガ口ースゲルなどを用いて電気泳動等によ つて分離し、ェチジゥムブ口マイ ド等によって染色することによって検出できる。 ②凝集性を有する酵母から調製したゲノムを鎵型として上記プライマ一を用い て PCR法によって核酸の増幅を行う。 得られた増幅産物を公知の方法にて簡易 精製した後に、 放射性元素あるいは蛍光色素等で標識する。 これをプローブとし て、 判定対象の酵母ゲノムを電気泳動した後にメンブラン上にブロッテイングし た試料とハイブリダィズさせるハイブリダイゼーシヨンを行う。 適用した標識方 法にあわせてハイブリダィズの状況を検出する。
実施例
以下に実施例を用いて本発明を具体的に説明するが、 本発明はこれに限定され るものではない。
実施例 1 : 下面ビール酵母の凝集^ έの判定
① 酵母 DNAの抽出
下面ビール酵母 30株をそれそれ 10mlの YPD培地 (l%Yeast Extract, 2% Peptone, 2%Dextrose) において定常期まで培養した後、 遠心分離により集菌し た。その後、蒸留水で洗浄、集菌をおこなつた。 0.2mlの 2% Triton X- ιΟΟ, 1% SDS, lOOmM NaCl, 10mM Tris-HCl(pH8.0), ImM Na2EDTAに懸濁した。 さらにフ ェノール · クロ口ホルム · イソアミルアルコール (25:24: 1) 0.2ml および酸洗浄 を行ったガラスビーズ 0.3g を加えて、 3分間十分に攪拌した。 TE(10mM Tris-HCl(pH7.5), ImM EDTA) 0.2ml を添加した後、 5分間遠心分離した。 水 槽を分離した後、 1mlの 100%エタノールを加えて、 10 分間以上、 — 20°Cに静 置した後、 再度遠心分離して沈殿を回収した。 得られた沈殿を前述の TE0.4ml に溶解させ、 10mg/ml RNAse A溶液を 3〃1加え、 37°C、 5分間加温した。 続 いて の 4M酢酸アンモニゥムおよび lmlの 100%エタノールを加えて、 10 分間以上、 — 20°Cに静置した後、 再度遠心分離して沈殿を回収した。 得られた 沈殿を 50 の蒸留水に溶解させ、 このうちの一部を試験に供した。
② 凝集性遺伝子 M 配列の一部を用いた PCR法による凝集性の判定 サッカロマイセス ·セレピシェ ( accharomy es cerevisiae) の第 VIII 染色 体上に存在する凝集性遺伝子 FL05の O F配列を基にブラィマー対を設計した。 プライマ一対の例を表 1 に示す。 PCR反応条件は、 94。C 2分間のホットス夕 —トの後、 94。C 1分間、 57.5°C 2分間、 72°C 2分間を 30サイクル繰り返し、 最後に 72°C 20分間保温した。 この一部を取り出し、 1%ァガロースゲルを用い た電気泳動に供した。 一方、 同じ菌株について YPD培地にて培養後、 麦汁培地 で発酵し、 先に記載した Bums法にて凝集性を評価した。本方法は酵母を凝集 · 沈降しやすい環境下 (すなわち pH4.5前後、 低温) で、 凝集促進物質である Ca イオンと共存させることにより、 人工的に凝集性を発現させてその凝集 ·沈降状 況を测定する方法であり、 沈降量を持って凝集性を判定する。 これらの結果の一 部、 プライマー aと cを対で用いて PCR反応により増幅した結果を図 1に示す が、 約 5,000bpの増幅 DNAの存否と Burns法にて評価した結果が一致してい ることを確認した。 本発明により、 下面ビール酵母の凝集性株 (ブルッフ酵母) と非凝集性株 (シュタウプ酵母) を判別することが可能であった。 すなわち、 本 発明により短時間に下面ビール酵母の凝集性を評価することが可能となり、 スク リ一二ングの際の一手法となりうることが確認された。

実施例 2 : 下面ビール酵母の凝集性変異株の評価
3種類の凝集性下面ビール酵母それそれに由来する凝集性株および自然変異に より生じた非凝集性株を実施例 1に示した方法にてそれそれ定常期まで培養した 後に、 DNA を抽出した。 同様に前掲表 1 に示すプライマ一を対で用いて PCR 反応を行わせ、 増幅産物を電気泳動に供した。
その結果の一部、 プライマー aと cを対で用いて PCR反応を行わせた時の結 果を図 2 に示すが、 約 5,000bpの増幅 DNAの存否により凝集性細胞と非凝集 性細胞を判別することができ、 本方法によって凝集性変異株を評価することが可 能であると確認された。 また、 ¾O,5配列両末端 (すなわちプライマー aおよび c) 以外の任意のブライマ一対によっても凝集性の有無を評価することが可能で あった。
実施例 3 : 酵母群中に占める変異株のポピュレーション調査
① シングルコロニーの単離および酵母 DNAの抽出
同一酵母に由来し、 ビール発酵中の凝集性の異なる酵母群 3群をそれそれ YPD 平板培地( l%Yeast Extract, 2% Peptone, 2% Dextrose, 2% Agarose) に塗布し、 25°C、 3日間培養して、 シングルコロニーを数十株ずつ単離した。 実施例 1記 載の方法でシングルコロニーそれそれの株を定常期まで培養した後、 集菌、 洗浄 し、 自動 DNA抽出装置 (東洋紡 Mag-Extractor Π) によって酵母の DNAを抽 出した。
② PCR法による凝集性の判定
上記記載の方法により同様に前掲表 1に示すプライマ一を対で用いて PCR反 応を行わせ、 増幅産物を電気泳動に供した。
その泳動結果の一部、 プライマ一 aおよび cを対で用いた結果を図 3 に示す 、 約 5,000bpの DNAの増幅の有無のみならず、 増幅するバンドサイズに差異 のある株の存在が認められた。 PCR による増幅状況の異なる株を 50ml の小ス ケールにて麦汁を用いた発酵試験に供した後に、 先に記載した Burns 法により 凝集性を評価した。 その結果、 約 5,000bpの DNAの増幅断片の有無と凝集性の 有無の間に一致が認められ、 さらに、 DNA増幅断片のサイズが短くなつた株で 凝集性の低下が認められた。 本方法により凝集性を失った株の検出のみならず、 これまで報告のない凝集性の低下あるいは消失の恐れのある株の検出が可能であ つた。
③ 変異株のポピュレーション調査
上記試験の結果、 表 2に示すように 3つの酵母群では凝集性株の存在割合が異 なっており、 凝集性の良い酵母群では凝集性株の存在割合が高く、 凝集性が非常 に低下した株では凝集性株の存在割合が極めて低下していることが確認された。 凝集性株の存在割合が低い酵母群では発酵中の浮遊酵母数がより多く、 発酵後の 酵母回収に困難をきたすことが予想され、 本方法を用いて実用上に影響する凝集 性変異株の存在割合を把握することが可能であった。
表 2 実施例 4 : サザンハイブリダイゼーシヨンにより下面ビール酵母の凝集性を 判定する方法 凝集性酵母に由来する凝集性株および自然変異により生じた^凝集性株を YPD培地で定常期まで培養した後、 全 DNAの抽出を Methods in Cell Biology (Academic Press Vol.12, p39-44, 1975)記載の方法にて行った。 抽出した DNA を EcoRIあるいは BamHIの制限酵素 (宝酒造製) により消ィ匕し、 1%ァガロー スゲルを用いて電気泳動した後にナイロンメンブランハイボンド N+ (アマシャ ム 'フアルマシア製) にブロッテイングし、 サザン解析に供した。
凝集性酵母 DNAを鋅型として前掲表 1 に示すプライマ一対をもちいて PCR 反応を行って増幅した産物を、 1%ァガロースゲルにて電気泳動し、 約 5,000bp の断片を紫外線照射下で切り出した後、 QIA quick Gel Extraction Kit (QIAGEN 社)によって抽出したものをプローブとし、 ECL検出システム (アマシャム ·フ アルマシア製) を用いてプロトコール通りに実施した。
プライマー aおよび cを対として PCR反応を行わせて得られた増幅断片をブ ローブとした場合の結果を図 4に示すが、 凝集性株に認められる糸 J 9.3Kbのバ ンドが非凝集性株では認められず、 本方法を用いて凝集性の判定を行うことが可 能であった。
産業上の利用の可能个生
本発明により、 判定対象の下面ビール酵母を発酵を行うことなく、 従来よりも 容易かつ迅速に高精度の凝集性有無の判定および非凝集性化変異の検出が可能に なり、 一度に多量のサンプルを扱えることから、 研究開発のみならず、 工業生産 における工程管理および品質管理にも有益である。

Claims

請求の範囲
1. 実験室酵母サヅカロマイセス 'セレピシェ {Sa charomv ^ cerevisiae) の 凝集性遺伝子 FLQ5の ORF配列を利用することを特徴とする下面ビール酵母の 凝集性有無の判定方法。
2. 実験室酵母サッカロマイセス 'セレピシェ {Saccharomvc.es cerevisiae) の 凝集性遺伝子 FLQ の ORF配列を利用することを特徴とする下面ビール酵母の 凝集性変異による非凝集性化または凝集性低下の有無の判定方法。
3. 実験室酵母サッカロマイセス 'セレピシェ ( Sa charomvn s cerevisiae) の 凝集性遺伝子 FL05の ORF配列を利用することを特徴とする下面ビール酵母の 凝集性変異による非凝集性化または凝集性低下を予知する方法。
4. サンプル酵母株を培養後、 DNAを抽出し、 凝集性遺伝子 ELQ5_の ORF配 列から設計した連続した 10 bp以上の塩基からなるプライマ一を用いて、 該 D N Aと P C R反応を行い、 増幅産物の有無を検出することを特徴とする請求項 1 記載の下面ビール酵母の凝集性有無の判定方法。
5. サンプル酵母株を培養後、 DNAを抽出し、 凝集性遺伝子 ELQ5_の ORF配 列から設計した連続した 10 bp以上の塩基からなるプライマ一を用いて、 該 D N Aと PC R反応を行い、 増幅産物の有無を検出することを特徴とする請求項 2 記載の下面ビール酵母の凝集性変異による非凝集性化または凝集性低下の有無の 判定方法。
6. サンプル酵母株を培養後、 DNAを抽出し、 凝集性遺伝子 ELQ5_の ORF配 列から設計した連続した 10 bp以上の塩基からなるプライマーを用いて、 該 D NAとPCR反応を行ぃ、 凝集性を示す酵母株の同増幅産物の DN Aのサイズと を比較することを特徴とする請求項 3記載の下面ビール酵母の凝集性変異による 非;疑集性化または凝集性低下の有無の予知方法。
7. 酵母群からシングルコロニーを数点単離し、 各コロニーごとに培養後、 DN Aを抽出し、 凝集性遺伝子 ELQ5_の ORF配列から設計した連続した 1 Obp以 上の塩基からなるプライマーを用いて、 該 DNAと PCR反応を行い、 増幅産物 の有無を検出し、 酵母群中の凝集性変異株のポピュレーシヨンを把握することを 特徴とする請求項 2記載の下面ビール酵母の凝集性変異による非凝集性化または 凝集性低下の有無の判定方法。
8 . 酵母群からシングルコロニーを数点単離し、 各コロニーごとに培養後、 D N Aを抽出し、 凝集性遺伝子 FLQ の ORF配列から設計した連続した 1 0 b p以 上の塩基からなるプライマーを用いて、 該 D N Aと P C R反応を行 、 増幅産物 の有無を検出し、 酵母群中の凝集性変異株のポピュレーションを把握することを 特徴とする請求項 3記載の下面ビール酵母の非凝集性化または凝集性低下を予知 する方法。
9 . 凝集性を示す下面ビール酵母から調製したゲノムを鎵型とし、 凝集性遺伝子 FLO の ORF配列から設計した連続した 1 0 b p以上の塩基からなるプライマ —を用いて PCR により増幅した産物をプローブとして、 サンプル酵母を培 #|麦 抽出した D N Aとハイブリダィゼーシヨンを行うことを特徴とする請求項 1記載 の下面ビール酵母の凝集性有無の判定方法。
1 0 . 凝集性を示す下面ビール酵母から調製したゲノムを錡型とし、 凝集性遺伝 子 FLO の ORF配列から設計した連続した 1 0 b p以上の塩基からなるブラィ マ一を用いて PCR により増幅した産物をプローブとして、 サンプル酵母を培養 後抽出した D N Aとハイブリダィゼ一シヨンを行うことを特徴とする請求項 2記 載の下面ビール酵母の凝集性変異による非凝集性化または凝集性低下の有無の判 定方法。
1 1 . 凝集性遺伝子 FLO の ORF配列から設計した連続した 1 0 b p以上の塩 基からなるプライマーの塩基配列が配列番号 1から 4のいずれかである請求項 4 から 9のいずれか 1項記載の方法。
1 2 . プライマ一セッ卜が配列番号 1または 2の第一のプライマーと配列番号 3 または 4の第二のブライマ一からなる請求項 1 1記載の方法。
